Application of Auto40 flow cytometry in immunodetection of HIV-infected patients

Abstract: Laboratory researchers from Yaoundé, the capital of Cameroon, have published two articles on HIV prevention: "Validation of a single-platform, volumetric, flow cytometry for CD4 T cell count monitoring in therapeutic mobile unit", and " Single-platform, volumetric, CD45-assisted pan-leucogating Auto flow cytometer for absolute and percent CD4 T cell counting in resource-constrained settings: Validation in Cameroon", they evaluated the mobile health center equipped with Apogee away from laboratory conditions The ability to achieve CD4+ T cell counts provides a new approach to AIDS prevention and treatment in remote areas.

The patient's immune system is attacked by HIV, the body's resistance will be reduced, and eventually the chance of replicating immune cells will be lost, and other diseases will lead to the death of various diseases. Therefore, in the clinical, real-time detection of the number of patients with CD4 + T cells will help to carry out targeted treatment in the treatment of AIDS. The most common method currently used is to count CD4 ⁺ T cells on a conventional flow cytometer under laboratory conditions. However, conventional flow cytometry is too large and expensive, and it cannot meet the needs of AIDS prevention and treatment in poor areas.

In the first literature study, the researchers evaluated the Auto40 flow cytometer (Apogee Flow Systems Ltd, Hemel Hempstead, UK) as an independent experimental platform for counting CD4 ⁺ T cells and laboratory flow with controls. A comparative study was performed on the cytometer FACSCalibur. 234 K3-EDTA blood samples (146 adults, 88 children samples) were counted and counted in CD4T cells using Auto40 and FACSCalibur. The results were statistically highly correlated (r(2)= 0.97 and r(2) = 0.98, respectively).

Another study used a mobile health center equipped with Apogee to conduct HIV screening activities in the suburb of Yaounde (screening at 20km in the control laboratory) and Ambang Bikok (53km from Yaounde). The two screenings included 188 volunteers (93 of whom were younger than 5 years old) and 69 adult volunteers. In the samples taken in the suburb of Yaounde, the mean CD4+ T cells in HIV/AIDS patients were 996 +/- 874 cells/μl using Auto40, and the results using FACSCalibur were 996 +/- 874 cells/μl; in Ambang Bikok, The mean CD4+ T cells of HIV/AIDS patients detected by Auto40 were 1041 +/- 317 cells/μl, and the results of the FACSCalibur test were 1032 +/- 294 cells/μl. The results of Yaounde (r2=0.982) and Ambang Bikok (r2=0.921) indicate that Auto40 is highly correlated with the results of FACSCalibur detection. Bland-Altman analysis shows that the results of Auto40 and FACSCalibur are very consistent.

Two studies have shown that the Auto40 flow cytometer installed in the Mobile Health Center enables HIV testing activities to be performed away from the laboratory, and the results are highly consistent with the controlled laboratory methods. The Auto40 flow cytometer can be used as a reliable device for monitoring the HIV infection status of adults and children under the conditions of WHO under normal conditions. This will facilitate the detection of HIV in HIV-infected areas in areas where laboratory equipment is lacking.
